Project title: Neuropathic pain treatment: Gene transfer to nociceptive neurons
Project summary: Neuropathic pain is a maladaptive, persistent pain syndrome that results from damage to the peripheral or central nervous system (CNS). Its pathogenesis is incompletely understood and treatments are often inadequate. Exploration of the mechanisms producing neuropathic pain has been aided by the interventions targeted to dorsal root ganglion (DRG) which is the primary site of initial pathological changes that lead to neuropathy. A strong impetus for developing a suitable model of DRG injection is the avoidance of systemic and CNS toxicity during neuropathic pain treatment. However, targeted DRG delivery in small animal subjects is a methodological challenge, as DRGs are enclosed within the intervertebral foramen and are therefore not easily accessible for injection. Our goal is to adopt a gene transfer technology targeted to DRG nociceptive neurons, and with the help of this methodology strengthen previous and gain new collaborations, increase our research and innovation capacities and attract international funding. Acquiring this technique, will additionally enable education of your scientists in our PhD programs.
Hrvatski sažetak: Neuropatska bol je oblik kronične boli koji nastaje kao rezultat ozljede perifernog ili središnjeg živčanog sustava. Njezina patogeneza nije dovoljno poznata, a načini liječenja su često neadekvatni. Istraživanje mehanizama neuropatske boli značajno je potpomognuto metodama ciljane primjene lijekova u spinalni ganglij koji je početno mjesto patoloških promjena koje dovode do neuropatske boli. Jak motiv za razvijanje tehnika ciljane primjene lijekova u spinalni ganglij je izbjegavanje sustavnih učinaka i neurotoksičnosti tijekom terapije neuropatske boli. Ciljana primjena lijekova u spinalni ganglij u malih eksperimentalnih životinja predstavlja priličan metodološki izazov zbog smještaja spinalnog ganglija u intervertebralnom foramenu. Naš cilj je usvojiti tehnologiju ciljanog prijenosa genskog materijala u nociceptivne neurone spinalnog ganglija. Pored toga nove tehnike omogućit će nam jačanje postojećih i stjecanje novih suradnji, povećati naše istraživačke i inovacijske kapacitete te privući financiranje projekata iz međunarodnih izvora. Usvajanje spomenute tehnike osigurat će i bolje obrazovanje mladih znanstvenika u okviru fakultetskih doktorskih studija.
